DOTMA Liposomes for DNA/RNA Delivery


DOTMA (N-[1-(2,3-dioleyloxy)propyl]-N,N,N-trimethylammonium) liposomes are a type of lipid-based delivery system used to transport DNA or RNA molecules into cells. These liposomes are positively charged due to the presence of the trimethylammonium group, which allows them to interact with negatively charged DNA or RNA molecules and facilitate their uptake by cells. The liposomes are made by mixing the lipid DOTMA with other lipids such as cholesterol, DOPE. This mixture is then hydrated in a buffer solution containing the DNA or RNA of interest, resulting in the formation of liposome-DNA or liposome-RNA complexes. The resulting complexes can be used to deliver DNA or RNA to cells both in vitro and in vivo.
