Diacylglycerol Pyrophosphate (DGPP)

Diacylglycerol pyrophosphate (DGPP) is a membrane phospholipid that has an important role in the regulation of intracellular signaling pathways.  DGPP is synthesized by the phosphorylation of diacylglycerol (DAG), which is released during the breakdown of phosphatidylinositol 4,5-bisphosphate (PIP2) by phospholipase C (PLC).  The enzyme responsible for the synthesis of DGPP is called DG kinase (DGK), and several isoforms of this enzyme have been identified in mammalian cells.  Once synthesized, DGPP can act as a second messenger to activate downstream signaling pathways, such as protein kinase C (PKC) and mitogen-activated protein kinase (MAPK) signaling cascades.  DGPP has also been shown to have a regulatory role in lipid metabolism, and alterations in DGPP levels have been linked to several diseases, including cancer and metabolic disorders.
