Phosphoinositides (PI) are minor components of cellular membranes found in eukaryotes. These molecules are similar to other phospholipids, but their head group is a cyclic myo-inositol. This inositol head group possesses free hydroxyl groups at positions D2-D6 and is readily phosphorylated by lipid kinases at hydroxyl groups D3, D4, and D5. The seven different combinatorially phosphorylated forms (PIPns) of the inositol head group contain valuable information content. PI molecules function as acidic addresses that identify different membranes, and they instruct proteins on how to behave in those membranes. These phosphorylated inositol groups can be found in various locations, including the endoplasmic reticulum, early and late endosomes, trans-Golgi, secretory granules, and the plasma membrane.
