Description

Myc is a family of proto-oncogenes that are essential transcription factors, belonging to the superfamily of basic helix-loop-helix (bHLH) DNA-binding proteins. C-myc (MYC), L-myc (MYCL) and N-myc (MYCN) are the three main members of the Myc family involved in cellular metabolism, cell division, differentiation and cell death. After forming a heterodimer with Myc-associated protein X (Max), Myc activates transcription by binding to E-box regions in the target gene promoters. c-Myc plays a fundamental role as a "double-edged sword" that promotes iPS cell generation and malignant transformation, while L-Myc promotes nuclear reprogramming by significantly downregulating the expression of differentiation-related genes. N-Myc-driven neuroendocrine tumors tend to highly express NEUROD1, resulting in enhanced metastatic potential. This product is designed as a tool for the delivery and expression of L-myc mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of L-myc mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The L-Myc protein is about 40 kD and consists of 364 amino acids |

Technical Notes
|
Work with mRNA-LNP on ice. It is important to minimize the time that the product spends at room temperature; after handling product during experiments, return immediately to ice.Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container; it is important not to spin for too long as this may rupture mRNA-LNP’s. Do not vortex. mRNA-LNP products should only be handled with certified RNAase-free reagents and consumables; use of filtered pipette tips is highly recommended |
