Description CD70 (Cluster of Differentiation 70) is a type II transmembrane glycoprotein encoded by the CD70 gene and belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF27/CD27 and a surface antigen on activated but not resting T and B lymphocytes. CD70 expression is upregulated after dendritic cell maturation. CD27 is the sole receptor for CD70, a member of the TNF receptor family that is constitutively expressed on thymocytes, naive T cells, B cells, and natural killer cells. Through binding to CD27, CD70 induces the proliferation of co-stimulatory T cells, enhances the generation of cytolytic T cells, and promotes T cell activation. This cytokine has also been reported to play a role in the regulation of B cell activation, the cytotoxic function of natural killer cells, and immunoglobulin synthesis. CD70 has been shown to be overexpressed in several types of cancer, including Hodgkin's and non-Hodgkin's lymphomas, as well as certain types of solid tumors. This overexpression is associated with cancer initiation and progression, suggesting that anti-CD70 antibodies may be a possible treatment for CD70-positive lymphomas. This product is designed as a tool for the delivery and expression of TF-tagged human CD70 mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of hCD70-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The hCD70-TF protein is about 23 kD, consisting of hCD70 and a C-terminal TF tag (15 amino acids)

Technical Notes Work with mRNA-LNP on ice. It is important to minimize the time that the product spends at room temperature; after handling product during experiments, return immediately to ice.Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container; it is important not to spin for too long as this may rupture mRNA-LNP’s. Do not vortex. mRNA-LNP products should only be handled with certified RNAase-free reagents and consumables; use of filtered pipette tips is highly recommended
