Description OX40L, also known as CD252 or TNFSF4, is a cytokine encoded by the TNFSF4 gene and belongs to the tumor necrosis factor ligand family. It is stably expressed on many antigen-presenting cells, such as type 2 conventional dendritic cells, macrophages, and activated B lymphocytes. Instead, its receptor, OX40, is found on the surface of activated T cells, natural killer cells, natural killer T cells and neutrophils. The OX40/OX40L signaling pathway plays complex roles in regulating T cell function. Under certain conditions, it strongly promotes the induction of Th1, Th2 and Th9 cells, while under other conditions it inhibits the generation of forkhead box p3 (Foxp3) Tregs6. Blockade of the OX40–OX40L pathway using OX40 or OX40L binders appears to suppress autoreactive T cells and improve autoimmune disease. However, enhancing OX40 signaling using OX40 agonist antibodies appears to enhance T cell-mediated antitumor immunity, contributing to tumor regression and prolonged survival. Thus, as a key immune checkpoint, activation or blockade of OX40-OX40L signaling could have therapeutic effects. This product is designed as a tool for the delivery and expression of TF-tagged human OX40L mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of hOX40L-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The hOX40L-TF protein is about 22 kD, consisting of hOX40L and a C-terminal TF tag (15 amino acids)

Technical Notes Work with mRNA-LNP on ice. It is important to minimize the time that the product spends at room temperature; after handling product during experiments, return immediately to ice.Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container; it is important not to spin for too long as this may rupture mRNA-LNP’s. Do not vortex. mRNA-LNP products should only be handled with certified RNAase-free reagents and consumables; use of filtered pipette tips is highly recommended
