Interleukin 12 (IL-12) is a p70 heterodimeric cytokine consisting of two subunits, IL-12alpha and IL-12beta, which are encoded by two separate genes, IL-12A (p35) and IL-12B (p40). IL-12 is expressed in dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells in response to antigenic stimulation. This cytokine is primarily responsible for the induction and enhancement of cell-mediated immunity and acts to induce TH1 cell differentiation, increase T cell and NK cell activation and cytotoxicity, and suppress or reprogram immunosuppressive cells. In preclinical studies, IL-12 has demonstrated significant antitumor effects against a range of malignancies. But severe toxicities in early clinical trials appear to dampen enthusiasm for IL-12-based immunotherapies. This product is designed as a tool for the delivery and expression of IL12B-FLAG-VPG-IL12A (secreted) mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of IL12B-FLAG-VPG-IL12A mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The IL12B-FLAG-VPG-IL12A fusion protein is approximately 56 kD, consists of full-length IL-12beta and mature IL-12alpha with a Flag tag (8 amino acids) and VPG peptide (27 amino acids) in between |

Work with mRNA-LNP on ice. It is important to minimize the time that the product spends at room temperature; after handling product during experiments, return immediately to ice.Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container; it is important not to spin for too long as this may rupture mRNA-LNP’s. Do not vortex. mRNA-LNP products should only be handled with certified RNAase-free reagents and consumables; use of filtered pipette tips is highly recommended |
