Description LAG-3 (lymphocyte activation gene 3) is encoded by the LAG3 gene. It is a member of the Ig superfamily and contains 4 extracellular Ig-like domains. LAG3 shares significant homology with CD4, but binds major histocompatibility complex class II with approximately 100-fold higher affinity than CD4. LAG3 is present on activated CD4+ and CD8+ T cells, CD4+ regulatory T cells, natural killer cells, and occasionally on immunosuppressive plasma cells. High levels of LAG3 on T cells are associated with a reduced ability to clear and control viruses such as HIV. In the context of chronic viral infection or tumors, LAG3 is frequently coexpressed with other immune checkpoint markers and defines significant T cell dysfunction. Dual blockade of the PD-1 axis using the immune checkpoint LAG3 is under clinical investigation. This product is designed as a tool for the delivery and expression of human TF tagged LAG-3 mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of LAG-3-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The LAG-3-TF fusion protein is approximately 60 kD, consisting of LAG-3 (525 amino acids) and a C-terminal TF tag (15 amino acids)

Technical Notes Work with mRNA-LNP on ice. It is important to minimize the time that the product spends at room temperature; after handling product during experiments, return immediately to ice.Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container; it is important not to spin for too long as this may rupture mRNA-LNP’s. Do not vortex. mRNA-LNP products should only be handled with certified RNAase-free reagents and consumables; use of filtered pipette tips is highly recommended
