Description VISTA (V-domain Ig suppressor of T cell activation), also called VSIR or B7-H5, is a protein belonging to the B7 family and encoded by the VSIR gene. This type I transmembrane protein has been found to function as an immune checkpoint, mainly expressed in myeloid cells such as macrophages, monocytes, dendritic cells, and T cells, and highly expressed in a variety of tumors, including melanoma, pancreatic cancer, prostate cancer, non-small cell lung cancer, and renal cell carcinoma. Blockade of VISTA by its blocking mAb enhances protective antitumor immunity and impaired the suppressive character of the tumor microenvironment (TME). VISTA significantly caused the inhibition of CD8+ T-cell activity in acute myeloid leukemia. B7-H5 expression has been found to be markedly increased in tumor tissues of colorectal cancer patients, and the increased VISTA expression is negatively correlated with CD8+ T-cell infiltration in CRC tissues. but the clinical significance of the B7-H5 expression in CRC and the role of VISTA in the TME remain to be elucidated. Such clinical studies are ongoing to explore the clinical significance of VISTA. This product is designed as a tool for the delivery and expression of human TF tagged VISTA mRNA for research. The product leverages the lipid nanoparticle (LNP) technology platform for simple and efficient delivery of VISTA-TF mRNA to a variety of mammalian cells in vitro and in vivo. The LNPs used are formulated with SM-102, DSPC, cholesterol and DMG-PEG2000 at an optimal molar concentration for a high rate of encapsulation and efficient mRNA delivery. The VISTA-TF fusion protein is approximately 36 kD, consisting of VISTA (311 amino acids) and a C-terminal TF tag (15 amino acids)

Technical Notes Work with mRNA-LNP on ice. It is important to minimize the time that the product spends at room temperature; after handling product during experiments, return immediately to ice.Upon receiving product, briefly pulse spin before opening to ensure product is at bottom of container; it is important not to spin for too long as this may rupture mRNA-LNP’s. Do not vortex. mRNA-LNP products should only be handled with certified RNAase-free reagents and consumables; use of filtered pipette tips is highly recommended
