Dendrimer-modified WS2 nanosheets as a pH and thermosensitive nanocarrier for capecitabine controlled delivery by near-infrared laser irradiation for breast cancer cell

Abstract

For the first time, a new type of pH/thermosensitive polymer was synthesized with allyl alcohol and N-vinyl-caprolactam on the tungsten disulfide surface. The dendrimer was grown from the surface of the modified tungsten disulfide as a nanocarrier to encapsulate the capecitabine. The obtained nanoparticles were charac-terized using different techniques. Various types of parameters were optimized. Drug release experiments were performed at different temperatures and pHs. To address these issues, a near-infrared laser irradiation drug delivery system was developed. The release rate of capecitabine from nanocarrier at pH 5.6 was remarkably higher than that at pH 7.4. The nanocarrier could release more than 90% of the drug at 45 degrees C in pH = 5.6 within 6 h. The release curves of drug from nanocarrier revealed strong dependences on near-infrared laser irradiation. This study aimed to evaluate the cytotoxicity of pH/thermosensitive nanocarrier through MTT assay against MCF-7 breast cancer and normal breast cells. The MTT assay indicated that the pH/thermosensitive nanocarrier significantly increased the rate of cell death in MCF-7 breast cancer cell line when compared to normal breast cells. The results revealed that pH/thermosensitive nanocarrier can be used as controlled drug delivery systems for breast cancer cells. The synthesized nanocarrier are a promising candidate for controlled release of capecitabine.
