Liposomal DOTA


DOTA-encapsulated liposomes are a vehicle that can be used to remotely load radioactive divalent ions, such as radioactive copper, into liposomes. A pH gradient is required when loading radioactive divalent ions. Divalent ions cross the liposome membrane when there is a proton flux across the lipid membrane when the pH is different between the interior and exterior of the liposome. Due to this flux, divalent cations move in opposite directions from the exterior to the interior of the liposome. Moreover, since the proton flow from the inside of the liposome to the outside of the liposome is constant, and the liposome is a thermodynamically stable structure, the permeability to divalent cations is not strong after formation. Without a pH gradient, divalent cations cannot move from the exterior to the interior of the liposome, they will remain on the exterior and will not cross the membrane.
